vectorette method
A method for PCR cloning an unknown sequence of DNA attached to a known sequence. To the end of the unknown sequence is attached a vectorette, a double-stranded sequence that includes a region of mismatch; primer for the known sequence drives the formation of a second primer site from the mismatch region and the unknown sequence is thus flanked with known sequences and can be specifically PCR amplified.
